EFFECTIVENESS OF ATONIC SOLUTION CONCENTRATION ON EXPLANT GROWTH OF GREEN BEANS (Vigna radiata L.) IN DROUGHT Stress CONDITIONS USING PEG 6000 IN VITRO ON MEDIUM Murashige and Skoog
Abstract
Atonik is an organic compound that is naturally synthesized by plants and influences physiological processes in plants as well as the auxin group which is in liquid form. This research aims to determine the effectiveness of atonic solution concentrations on the growth of green bean (Vigna radiata L.) explants in vitro under drought stress conditions, which are induced by PEG 6000 at different concentrations, as an effort to control drought stress in the growth process. This research was carried out in March - April 2023 in the Plant Tissue Culture Room, Botany Laboratory, Biology Department, Faculty of Mathematics and Natural Sciences, University of Lampung. This research design was prepared using the basic pattern of a 3x3 Factorial Design with two factors; The first factor is atonic solution with three concentration levels, namely 0 ml/l (A0), 1 ml/l (A1), 2 ml/l (A2) and the second factor is PEG 6000 w/v with three concentration levels, namely 0% ( B0), 3% (B1), 4% (B2) with 4 repetitions. Quantitative data was homogenized using Levene's test then analyzed using Analysis of Variance (ANOVA) at the 5% level, then if significant differences were obtained then the data was continued with the Tukey test at the 5% level. The results showed that concentrations of atonic solution of 1 ml/l and 2 ml/l with 3% and 4% PEG could increase explant growth.
